Many of us often think with dismay that ______ knows what we are going through.

a. nobody
b. none

Are they both correct?
If not, could you explain some?

Ismael's right, Kis, that only a works in this sentence.

The reason that b doesn't work is simply that it's a substitute pronoun, but since we have no idea what it's substituting for, it doesn't make sense.

Remember that none really is a contraction of "not one," so the logical question in this case would be, "Not one what?"
