If you begin a sentence with "unfortunately" or "usually," do you always need to put a comma after it?

No, you don't. It all depends on what follows those words. For example, if I continue with a preposition, I won't use that comma:

Unfortunately for the defendant, the judge decided to fine him $500.

Usually in cases like that, the judge will find against the defendant.

But if the sentence has a subject directly after those two words, we will need to use that comma:

Unfortunately, the defendant was fined $500.

Usually, the judge will find against the defendant in cases like that.
