I would like to know which one is correct:

1- In a follow up to our conversation.

2- In follow up to our conversation.

"Follow up" can act as a verb or as a noun.

In your phrases, "follow up" is a noun and it is a singular count noun. Therefore, it needs an article or other determiner. The correct phrase is:

In a follow up to our conversation...
_______

Other ways you might use "follow up" in an introductory phrase are

As a follow up to our conversation...
Following up on our conversation...
To follow up on our conversation...
