Are both sentences correct?
1.Peter runs fastest in the class.
2.1.Peter runs the fastest in the class.

You need something more in the sentence to make the superlative "fastest" work. You can say

"” Peter runs fastest of anyone in the class

"” Peter runs the fastest of anyone in the class

"” Peter is the fastest runner of anyone in the class
